Dosage information

CJC-1295 with DAC dosage

No human dose of CJC-1295 with DAC has been established for any use, and no form of it is part of an approved drug.13 Published studies gave healthy adults one to three injections under the skin, dosed at 20 to 250 micrograms per kilogram of body weight.45 These figures describe research, not advice.

Is there an established CJC-1295 dosage?

No. CJC-1295 with DAC has no approved use, so no label sets a dose, a schedule or a route.1 FDA found no approved product containing any form of it in the 12 other countries it checked, and none authorized in the European Union.2 Professional society guidelines do not mention it for diagnosis or treatment.3

What exists instead is a short record of research doses. The published studies were small and short and were run in healthy adults, and the one patient trial was terminated.219 Each figure below carries its species, route and schedule, so it can be read in context. None of them is a dose for a person.

Doses in human studies

The two Teichman trials lasted 28 and 49 days and were randomized, placebo-controlled and double-blind.4 Every published study that names a route gave CJC-1295 with DAC by injection under the skin.47

Study Who took part Route Doses tested Injections
Teichman 2006, single-dose trial 42 healthy adults aged 21 to 61: 35 on the drug, 7 on placebo48 Under the skin4 30, 60, 125 or 250 mcg/kg458 One
Teichman 2006, repeated-dose trial 24 healthy adults, 1 on placebo in each group of 65 Under the skin4 30 or 60 mcg/kg on days 0 and 14; 20 or 30 mcg/kg on days 0, 7 and 145 Two or three
Ionescu and Frohman 2006 12 healthy men aged 20 to 4067 Under the skin7 60 mcg/kg (4 men) or 90 mcg/kg (8 men)7 One, with overnight GH sampling before and 1 week after6
Sackmann-Sala 2009 11 healthy young men, a subset of the Ionescu and Frohman group2011 Not stated separately 60 to 90 mcg/kg11 One, with blood taken before and 1 week after20
ConjuChem phase 2, NCT00267527 (terminated) Adults aged 18 to 65 with HIV-associated visceral obesity: 120 in the registry, 192 in reports cited by FDA98 Not stated in the sources Weekly doses rising over 3 weeks through 60, 90 and 120 mcg/kg (low) or 60, 120 and 240 mcg/kg (high), then 9 more weeks; or placebo8 Weekly; 12 weeks in the registry9

What changed with the dose

  • Size of the hormone rise. How far GH and IGF-I rose after one injection depended on the dose, and IGF-1 stayed raised for at least 2 weeks at the two highest doses.45
  • Tolerability. The trial authors described CJC-1295 as relatively well tolerated, particularly at 30 or 60 mcg/kg.4 All side effects except short-lived hives at the injection site were more common at 125 and 250 mcg/kg.10 Diarrhea, for example, affected 45% of people at 125 mcg/kg and every person at 250 mcg/kg.10
  • Flushing. In the repeated-dose trial, the incidence of flushing was 40% after low-dose injections and 100% after high-dose injections.10
  • Heart rate. In the 12-man study, the rise in heart rate grew with the dose.11
  • Repeated injections. IGF-I effects built up from one injection to the next: peak IGF-1 after later injections was progressively higher than after the first.45

Every reaction the trials recorded is listed on the side effects page linked below.

Doses in animal studies

Species Route Dose and schedule Length Result relevant to dosing
Normal male Sprague Dawley rats Under the skin Amount and number of injections not given in the abstract 2-hour GH test; plasma levels followed past 72 hours 4 times the GH exposure of unmodified hGRF(1-29) over 2 hours; still present in plasma beyond 72 hours12
GHRH knockout mice, treated from 1 week of age Not given in the abstract 2 micrograms per mouse every 24, 48 or 72 hours 5 weeks Dosing every 24 hours gave normal weight and length; longer gaps improved growth without normalizing it13
Rats and dogs, in FDA's toxicology review Under the skin Repeated daily injections of 0.25 mg/kg or more14 Up to 14 days14 Bleeding, inflammation and tissue death at the injection site14

Animal doses were set per mouse or per kilogram of the animal, for each experiment's own purpose. They do not convert into an amount for a person.

Why the study doses are not a dose for anyone

  • Who was studied. All published dosing data come from healthy volunteers, 87% of them men.15 FDA found no study in people with a disease or condition.16
  • For how long. Most participants (73%) received one injection, and FDA calls the long-term safety of CJC-1295 in people unknown.15
  • The longest dosing ended early. In the phase 2 trial, a participant had a heart attack 2 hours after an 11th weekly dose and died about an hour later. The study was terminated, and its data have not been published.19
  • Which molecule. FDA says results for the DAC form cannot be applied to CJC-1295 without DAC.17 It also calls inconsistent naming of CJC-1295 substances a safety risk, because a patient may be dosed with a different substance than the one a physician ordered.18
  • How doses were set. Each published human dose was calculated from body weight in micrograms per kilogram, not given as a fixed amount.457
  • What the doses did not show. The studies did not measure outcomes such as lean body mass or fat mass, so FDA says they do not support effectiveness for growth hormone deficiency.3

Frequently asked questions

What is the dosage of CJC-1295 with DAC?

No dose has been established for any use in people. CJC-1295 with DAC is not FDA-approved, and FDA found no study that gave any form of it to people with a disease or condition.116 Published studies gave healthy adults 20 to 250 micrograms per kilogram of body weight by injection under the skin, once or up to three times.45 Those numbers record how research was designed. They are not guidance for anyone.

How often was CJC-1295 with DAC given in studies?

Usually once. Of the 63 people given it in published studies, 46 (73%) received a single injection.15 In the repeated-dose trial, groups were injected on days 0 and 14, or on days 0, 7 and 14.5 The terminated phase 2 trial was designed around weekly injections for 12 weeks.89 No published study followed people for longer than 49 days, and FDA calls the long-term safety of CJC-1295 in people unknown.415

How long does one injection of CJC-1295 with DAC last?

In healthy adults, the estimated half-life was 5.8 to 8.1 days.4 After a single injection, mean GH rose 2- to 10-fold for 6 days or more, and IGF-I rose 1.5- to 3-fold for 9 to 11 days, depending on the dose.4 When two or three injections were given a week or two apart, IGF-I stayed above baseline for up to 28 days.45 In rats, the peptide was still in plasma beyond 72 hours.12

Does CJC-1295 raise IGF-1 above normal?

Only at the highest dose tested. In the single-dose trial, IGF-1 went above the normal range for age and sex only in people given 250 mcg/kg.5 In 12 men given 60 or 90 mcg/kg, IGF-I was 45% higher a week later but stayed within normal limits in every man.67 FDA notes that approved growth hormone labels warn of risks tied to GH and IGF-1, such as glucose intolerance, and it has no data showing CJC-1295 would avoid them.11
