Kisspeptin-10 side effects

Kisspeptin-10 has no reported side effects in its small safety record: three studies of 4 to 13 people each found no kisspeptin-related adverse events.12253 With continuous exposure, kisspeptins and their analogs turned the reproductive hormone axis down in human and animal studies.456 FDA says it lacks the safety information to judge harm.7

Reported in kisspeptin-10 studies

  • No kisspeptin-related adverse events (human trials, 2011 to 2013): none were reported in 6 healthy men given single IV doses, in 13 healthy men followed for 2 weeks after one IV dose, or in 4 patients given 12-hour infusions.12253
  • No significant acute or chronic adverse effects: this is how the authors of a 2015 head-to-head trial in healthy men summarized kisspeptin's record in people.16
  • Short-term data only: no kisspeptin-10 study gave it for longer than a 22.5-hour infusion.26

Reported with kisspeptin-54 and synthetic analogs

These come from related compounds, not from kisspeptin-10, and are the closest data available.

  • No side effects (randomized trials, 32 men and 32 women with low sexual desire): 75-minute kisspeptin-54 infusions were well tolerated.89
  • No adverse effects over 8 weeks (human trial, women with hypothalamic amenorrhea): twice-weekly kisspeptin-54 injections.17
  • IVF complications (human trial, 53 women, 2014): five adverse events were established complications of IVF treatment and pregnancy: two ectopic pregnancies, one heterotopic pregnancy and two miscarriages.18
  • Mild OHSS (phase 2 trial, 60 women at high risk, 2015): mild early OHSS in 3 women (5%) and mild late OHSS in 1 (2%), with no moderate, severe or critical cases.1314
  • Injection-site reactions (human trial, 60 healthy men): with the synthetic analog TAK-683 given under the skin, these were the most common drug-related events, including redness in 6 men and pain in 3. After single doses, overall adverse event rates were similar with TAK-683 and placebo (31% and 28%).527
  • Mild to moderate events (phase 1 trials, 112 healthy men): grade 1 to 2 adverse events occurred in 26% of subjects during treatment with the analog TAK-448.2810
  • No serious adverse effects (human study, 24 women, 2020): after injection of MVT-602 or kisspeptin-54.19

Effects of repeated or continuous exposure

Across human and animal studies, steady exposure turned the reproductive axis down.45620

  • Fading response (human trial, 10 women with hypothalamic amenorrhea, 5 given kisspeptin-54 and 5 saline): injections were twice daily under the skin for 2 weeks.29 The LH rise after kisspeptin-54 fell from 24.0 IU/L on day 1 to 2.5 IU/L on day 14.4
  • Testosterone suppression (phase 1 trials, healthy men): with continuous TAK-683 or TAK-448 at the higher doses tested, testosterone fell into the castration range by day 7 to 8.510
  • LH pulse suppression (animal study, adult male rhesus monkeys): continuous high-dose IV kisspeptin-10 markedly suppressed the size and frequency of LH pulses.6
  • Testicular damage (animal study, adult male rats): 13 days of continuous kisspeptin-54 under the skin lowered testicular weight and caused degeneration of the seminiferous tubules.11
  • Dose matters (2014 review): the authors judged that high doses of kisspeptin-54 or kisspeptin-10 may desensitize the axis, and lower doses do not.20

Warnings

  • Not an approved drug: no kisspeptin product has FDA approval, and DailyMed holds no US label for one.12
  • FDA compounding list: Kisspeptin-10 has been in 503A Category 2, bulk substances that raise significant safety risks, since September 29, 2023, and remained there on the May 14, 2026 list.2421
  • Product quality: trial peptide was GMP-made and tested before use, 97% pure by HPLC in one study and checked for sterility and pyrogens in another,3031 while FDA flags impurity and characterization concerns for kisspeptin-10.24
  • Longest exposure in the studies reviewed: 8 weeks of twice-weekly kisspeptin-54 in women with hypothalamic amenorrhea.17
  • Athletes: the WADA 2026 Prohibited List bans kisspeptin for males at all times under S2.2.1.22

Frequently asked questions

What are the side effects of kisspeptin?

In the small kisspeptin-10 studies that reported safety, no kisspeptin-related adverse events occurred, with 4 to 13 people in each.12253 Kisspeptin-54 trials also reported good tolerability.89 With long-acting analogs, the most common drug-related problems were injection-site redness and pain, and continuous dosing lowered testosterone.510 FDA says it does not have the safety data to know if kisspeptin-10 would harm people, so long-term risks are unknown.7

What happens if you take too much kisspeptin?

In studies, more was not better. In healthy men, the top kisspeptin-10 dose tested (3.0 μg/kg) produced no significant LH rise, though lower doses did.123 Twice-daily kisspeptin-54 injections lost most of their LH effect within 2 weeks in women with hypothalamic amenorrhea.4 With continuous exposure, a high IV dose suppressed LH pulses in monkeys, and rats given kisspeptin-54 for 13 days developed degeneration of the sperm-producing tubules in the testes.611

Is kisspeptin-10 safe?

Its safety has not been established. The kisspeptin-10 studies that reported safety enrolled 4 to 13 people and gave it as single doses or for up to 12 hours, with follow-up of up to 2 weeks.12253 FDA lists Kisspeptin-10 among compounding substances that may present significant safety risks, citing possible immune reactions and impurity problems, and says it lacks the information to know whether it would harm people.247 It is not an FDA-approved drug.12

Does kisspeptin cause ovarian hyperstimulation syndrome (OHSS)?

In a 2015 IVF trial of kisspeptin-54 in 60 women at high risk of OHSS, mild early OHSS occurred in 3 (5%) and mild late OHSS in 1 (2%), and none had moderate, severe or critical OHSS.1314 In a later trial, a second dose 10 hours after the first did not add moderate OHSS: 1 of 31 women after one dose and 0 of 31 after two.15
