Dosage information

Kisspeptin-10 dosage

No human dose of kisspeptin-10 is established: there is no FDA-approved kisspeptin product and no label.1 Published doses come from small clinical studies that dosed it by body weight, mostly into a vein, as single injections or infusions lasting 90 minutes to 22.5 hours.2263 These study doses are not guidance for people.

No established human dose

No human dose of kisspeptin-10 has been established. There is no FDA-approved kisspeptin product, so there is no labeled dose,1 and FDA has said its information is too limited to tell whether kisspeptin-10 would cause harm in humans.9 The human figures below come from small, supervised research studies, and the US studies ran under FDA oversight.1011 The figures show what was tested, not what anyone should use.

Doses used in kisspeptin-10 studies

All of these studies were small, and none gave kisspeptin-10 for longer than 22.5 hours.26

Study People Route and dose How long Main result
George, 2011 6 healthy men IV bolus of 0.01, 0.03, 0.1, 0.3, 1.0 and 3.0 μg/kg, each against vehicle2 Single doses LH peaked by 45 minutes; no significant rise at 3.0 μg/kg29
George, 2011 4 healthy men IV infusion, 4 μg/kg per hour26 22.5 hours LH rose; testosterone rose from 16.6 to 24.0 nmol/L26
George, 2011 4 healthy men IV infusion, 1.5 μg/kg per hour31 9 hours LH pulse frequency rose from 0.7 to 1.0 per hour31
Jayasena, 2011 11 healthy men IV bolus of 0.3, 1.0, 3.0 or 10 nmol/kg32 Single doses Testosterone rose only after 0.3 and 1.0 nmol/kg28
Jayasena, 2011 35 healthy women IV bolus up to 10 nmol/kg; bolus under the skin up to 32 nmol/kg; IV infusion starting at 20 to 720 pmol/kg per minute323 Single doses; 90-minute infusions No significant change in the follicular phase by any route; LH and FSH rose in the preovulatory phase after 10 nmol/kg IV2827
Chan, 2011 13 healthy men IV bolus of 0.24 nmol/kg (0.313 μg/kg)33 Single dose Reset the timing of GnRH pulses33
Young, 2013 4 patients with NKB-pathway defects IV infusion, 1.5 μg/kg per hour12 12 hours LH pulses restored in all 412
George, 2013 5 men with type 2 diabetes and low testosterone IV infusion, 4 μg/kg per hour13 11 hours Testosterone rose from 8.5 to 11.4 nmol/L13
Jayasena, 2015 Healthy men, 5 per dose group IV infusion of 0.1, 0.3 or 1.0 nmol/kg per hour14 3 hours Similar to kisspeptin-54; GnRH gave about 3 times more LH and FSH3435
Chan, 2020 16 children (3 girls, 13 boys) with delayed or stalled puberty One IV dose of 0.313 μg/kg, under investigational new drug 113 59111 Single dose All 8 with an LH rise of 0.8 mIU/mL or more later progressed through puberty15

Reading the units

Doses were set by body weight, in micrograms (μg) or nanomoles (nmol) per kilogram. The two units convert through the molar mass, 1302.4 g/mol for kisspeptin-10,16 so 1 nmol weighs about 1.3 μg. That is why the 0.24 nmol/kg dose in the 2011 Massachusetts study equals the 0.313 μg/kg the authors also reported.33

Doses used for kisspeptin-54, a different peptide

The trials that draw the most attention used kisspeptin-54, which stays in the blood far longer, with a half-life of 27.6 minutes against about 4.36 These doses are listed for comparison only:

  • IVF egg maturation (2014 to 2017, including phase 2 trials): one injection under the skin of 1.6 to 12.8 nmol/kg. The 2015 and 2017 trials gave it 36 hours before egg collection, and the 2017 trial tested a second 9.6 nmol/kg dose 10 hours after the first.1718619
  • Low sexual desire and brain imaging: an IV infusion of 1 nmol/kg per hour for 75 minutes, on one study visit, compared with placebo.202122
  • Hypothalamic amenorrhea: 6.4 nmol/kg under the skin twice daily for 2 weeks, after which the response had faded, and twice-weekly injections for 8 weeks, with only partial desensitization.3945

Animal doses

These were given to animals and are not human doses:

  • Adult male rhesus monkeys: continuous IV kisspeptin-10, studied as metastin 45-54, at 200 or 400 μg per hour. The high dose markedly suppressed LH pulses.23
  • Adult male rats: kisspeptin-54 at 50 nmol per day under the skin for 13 days lowered testicular weight.24 A 1-day course raised LH and testosterone, but the effect was lost after 2 days.40

Why study doses are not a guide

  • Route: researchers state that kisspeptin-54, but not kisspeptin-10, can be given under the skin because of its longer half-life, and injections of kisspeptin-10 under the skin did not change reproductive hormones in follicular-phase women.3627
  • More was not stronger: in healthy men, the highest IV bolus tested gave no significant LH rise, while lower doses did.29
  • High doses can blunt the response: a 2014 review concluded that high doses of kisspeptin-54 and kisspeptin-10 may cause desensitization, while lower doses do not.25
  • Sex and cycle phase: the same IV doses raised LH in men but did not significantly change gonadotropins in follicular-phase women.2827
  • Material: trial peptide was made to GMP standards and tested before use, and FDA's stated concerns for kisspeptin-10 include impurities and characterization.371038

Frequently asked questions

How long should you take kisspeptin for?

No course of use has been established. Kisspeptin-10 studies gave single doses or IV infusions that lasted at most 22.5 hours.26 With the longer-acting kisspeptin-54, twice-daily injections lost most of their effect within 2 weeks, while twice-weekly injections over 8 weeks caused only partial desensitization in women with hypothalamic amenorrhea.45 These study lengths describe research conditions, not a recommended cycle.

When should I take kisspeptin-10?

There is no approved use and no established timing for kisspeptin-10.1 In research, timing changed the result: IV kisspeptin-10 raised LH and FSH in women in the preovulatory phase of the cycle but not in the follicular phase at the doses tested.2728 In the 2015 and 2017 IVF trials, kisspeptin-54 was given 36 hours before egg collection as part of supervised fertility treatment.619 Questions about any treatment belong with a licensed clinician.

How quickly does kisspeptin work?

Hormone changes start fast. After an IV kisspeptin-10 dose, LH rose rapidly in healthy men and peaked within 45 minutes.729 After an under-the-skin kisspeptin-54 injection in women having IVF, LH peaked 4 to 6 hours later.8 Kisspeptin-10 itself leaves the blood quickly: its plasma half-life was 3.8 minutes in men and 4.1 minutes in women.30
